Androgens downregulate BMP2 impairing the inductive role of dermal papilla cells on hair follicle stem cells differentiation

چکیده
Hair follicle cyclical regeneration is regulated by epithelial-mesenchymal interactions. During androgenetic alopecia (AGA), hair stem cells (HFSC) differentiation impaired deregulation of dermal papilla (DPC) secreted factors. We analyzed androgen influence on BMPs expression in DPC and their effect HFSC to lineage. Androgens downregulated BMP2 BMP4 spheroids. Addition restored alkaline phosphatase activity, marker hair-inductivity DPC, DPC-induced differentiation, both inhibited androgens. Concomitantly, differentiating HFSC, an upregulation BMPRIa BMPRII receptors nuclear β-catenin accumulation, indicative Wnt/β-catenin pathway activation, were detected. Our results present as androgen-downregulated paracrine factor that contributes inductivity favors lineage, possibly through a crosstalk with pathway. A comprehensive understanding androgen-deregulated factors effects would help improve treatments for AGA.

Purpose: The aim of this study was to evaluate the effect of NT-3 on the decrease of the differentiation time of bulge stem cells of rat hair follicle from neuron like cells.Materials and Methods: The bulge region of the rat whisker was isolated from and cultured in DMEM/F12 supplemented with epidermal growth factor (EGF) in 3 groups for 7, 8 and 9 days .Then 10 ng/ml NT-3 was added to each gro...
متن کاملSingle transcription factor reprogramming of hair follicle dermal papilla cells to induced pluripotent stem cells.
Reprogramming patient-specific somatic cells into induced pluripotent stem (iPS) cells has great potential to develop feasible regenerative therapies. However, several issues need to be resolved such as ease, efficiency, and safety of generation of iPS cells. Many different cell types have been reprogrammed, most conveniently even peripheral blood mononuclear cells. However, they typically requ...

متن کاملBMP signaling in dermal papilla cells is required for their hair follicle-inductive properties.
Hair follicle (HF) formation is initiated when epithelial stem cells receive cues from specialized mesenchymal dermal papilla (DP) cells. In culture, DP cells lose their HF-inducing properties, but during hair growth in vivo, they reside within the HF bulb and instruct surrounding epithelial progenitors to orchestrate the complex hair differentiation program. To gain insights into the molecular...
